The BRCA 1 Associated Protein 1 (BAP1) protein belongs to the ubiquitin C-terminal hydrolase subfamily of de-ubiquitination enzymes that are involved in the removal of ubiquitin from proteins. The encoded enzyme binds to the breast cancer type 1 susceptibility protein (BRCA1) via the RING finger domain of the latter and acts as a tumor suppressor. In addition, the enzyme may be involved in regulation of transcription, regulation of cell cycle and growth, response to DNA damage and chromatin dynamics. Germ line mutations in this gene may be associated with tumor predisposition syndrome (TPDS), which involves increased risk of cancers including malignant mesothelioma, Uveal melanoma and cutaneous melanoma. Cadherin 17, also known as LI cadherin (for liver-intestine cadherin), is a member of the cadherin family of calcium-dependent adhesion molecules. Cadherins mediate cell-cell interactions via hemophilic binding, and are critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Cadherin 17 expression is restricted to liver and intestine tissues and is specifically localized to the basolateral domain of hepatocytes and enterocytes. NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. Furthermore, NKX3. 1 shows better sensitivity than Prostate Specific Antigen (PSA) for identifying metastatic prostatic adenocarcinoma. This suggests that immunohistochemical staining of NKX3. 1, along with other prostate-restricted markers, may be valuable for the definitive determination of prostatic origin in poorly differentiated metastatic carcinomas. S100A and S100B proteins are two members of the S100 family of calcium binding proteins. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the beta-chain (i.e. in S-100A and S-100B) but not on the alpha-chain of S-100 (i.e. in S-100A and S100A0). This antibody can be used to localize S-100A and S-100B in various tissue sections. S-100 protein has been found in normal melanocytes, Langerhans cells, histiocytes, chondrocytes, lipocytes, skeletal and cardiac muscle, Schwann cells, epithelial and myoepithelial cells of the breast, salivary and sweat glands, as well as in glial cells. Defects in TNFRSF13C are the cause of immunodeficiency common variable type 4 (CVID4) ; also called antibody deficiency due to BAFFR defect. This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation. A significant overexpression of Aurora B has been found in a variety of human tumors including non-small cell lung carcinoma, astrocytoma, seminoma and carcinomas of the colon, prostate, endometrium and thyroid. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues.
